- Las enfermedades raras, poco frecuentes o poco prevalentes, son unas afecciones complejas que presentan importantes desafíos, tanto para el conocimiento científico actual como para la sociedad en su conjunto.

- Se trata en general de enfermedades crónicas y degenerativas que comienzan a manifestarse en los primeros años de vida. Además, se suelen manifestar como síndromes, (afecciones que comparten los mismos síntomas y que afectan a distintas partes del cuerpo de forma simultánea) lo que dificulta el diagnóstico. Quien la tiene la padece toda su vida y empeora con el transcurso de la misma, generando discapacidad motora, sensorial o intelectual. En una parte muy significativa de los casos, la enfermedad presenta riesgo de muerte, sobre todo en el rango de edad que comprende de los 0 a los 5 años, suponiendo una auténtica tragedia para las familias.
- Estas enfermedades también se conocen como «enfermedades huérfanas», debido a que, por su baja prevalencia, presentan una baja rentabilidad económica a la industria farmacéutica y difícilmente encuentran financiación para su investigación científica, lo que supone un verdadero drama para los afectados y las familias que buscan un diagnóstico para la enfermedad. Este difícil camino que sufren las personas afectadas en busca de soluciones médicas se conoce como «periplo diagnóstico».
